Abstract
Bioactive compound content in vitro culture can be improved by feeding precursor, The effect of
tryptophan on growth and catharanthine production in cell aggregates of Catharanthus roseus (1.) G
Don as well as in medium has been studied Agregate cells of C. roseus were cultured in Murashige
and Skoog (MS) medium supplemented with 2 mg/L NAA and 0,2 mg/L kinetin. Agregate cells culture
were divided into control and treated groups. The treated group u/as fed by 150 mg/L tryptophan while
the control was aciciecj by ciestilied water. The growth of cells and catharanthine production in the cell
aggregates as well as rn medium were observed on day 0, 4,7, 10. 14, and 21 Qualitative and
quantitative analysis of catharanthine were carried out using High Performance Liquid Chromatography
(HPLC). The resu[ showed that the grcMh pattern in the contrcl and treated cell aggregates were
similar, except additioned oi tryptophan coulci increase the growih of the cell aggregates, producing
longer growth phase. The addition of 150 mg/L tryptophan also increased catharanthin production. The
highest concentration of catharanthine was produced in cellaggregales2261,6Bt17,05 pg/g DW on the
