Abstract

Alga laut Eucheuma cotonii dan Eucheuma spinosum adalah spesies golongan alga merah yang digunakan sebagai bahan pangan sumber karagenan dan antioksidan. Senyawa antioksidan sangat dibutuhkan tubuh, karena berperan dalam menghambat terbentuknya radikal bebas yang menjadi sumber berbagai penyakit. Kedua spesies alga laut ini kaya akan senyawa bioaktif, sehingga banyak dibudidaya untuk memenuhi permintaan pasar. Senyawa antioksidan sebagai komponen metabolit sekunder, memiliki aktifitas berbeda-beda. Selama ini penelitian terhadap alga laut ini, masih terbatas pada kandungan karagenan dan pigmen, baik untuk industri maupun ekspor. Ekstraksi senyawa antioksidan dilakukan dengan cara maserasi menggunakan pelarut metanol pada beberapa konsentrasi. lsolasi senyawa dilakukan melalui kombinasi teknik pemisahan Kolom Kromatografi (KK), dan Kromatografi Lapis Tipis(Kl T). Karakterisasi senyawa dilakukan dengan NMR. Tujuan penelitian ini, adalah : 1) mendapatkan konsentrasi pelarut metanol yang tepat, untuk menghasilkan aktifrtas antioksidan terbaik pada ekstrak alga Eucheuma cotonii dan Eucheuma spinosum segar maupun kering, 2) mendapatkan hasil fraksinasi bertingkat dengan jenis pelarut terbaik, yang menghasilkan senyawa antioksidan dengan aktifrtas antioksidan terbaik, 3) mendapatkan senyawa yang mempunyai aktifrtas antioksidan dari ekstrak alga laut terpilih melalui isolasi dengan KK, Kl T, dan elusidasi struktur senyawa dengan NMR, menggunakan teknik H-NMR, C-NMR, HMQC, dan HMBC. Penelitian dilakukan dalam tiga tahap, yaitu : (I) Ekstraksi Senyawa Antioksidan dari Alga Eucheuma cotonii dan Eucheuma spinosum dalam kondisi Segar dan Kering Menggunakan Pelarut Metanol pada konsentrasi Berbeda (60, 70, dan 80%) dengan uji Kadar air, Total Fenol, DPPH, FRAP, dan Total Karotenoid, (II) Pengujian Fitokimia dan Aktifrtas Antioksidan Alga Eucheuma sp. terpilih Melalui Fraksinasi Bertingkat Menggunakan Pelarut Heksan, Etil Asetat, Butanol, dengan Uji Total Fenol, DPPH, FRAP, dan Total Karotenoid, (Ill) lsolasi viii dan Karakterisasi melalui Penentuan Struktur Senyawa Antioksidan Alga Eucheuma sp. secara In Vitro dengan KK, KL T, dan NMR. Hasil Penelitian menunjukkan Kandungan total fenol, DPPH, dan FRAP tertinggi hasil ekstraksi menggunakan pelarut metanol pada alga laut Eucheuma cotonii dan E. spinosum (dalam bentuk segar dan kering) dihasilkan oleh ekstrak metanol 60 % pada E. Spinosum segar. Kandungan Total Karotenoid tertinggi (sebesar 9,40 ± 0,35 μg/g} ditemukan pada sampel ekstrak metanol 60% untuk Eucheuma cotonii segar, dikuti oleh ekstrak metanol 60% untuk E. spinosum (8,73 ± 0,23 μg/g). Hasil analisis untuk keempat parameter yang diuji, Eucheuma spinosum segar pada ekstrak metanol 60% menunjukkan aktifitas antioksidan yang lebih baik dibandingkan dengan sampel lainnya. Sampel ini digunakan dalam penelitian tahap selanjutnya. Hasil pengujian fitokimia menunjukkan bahwa pada setiap ekstrak fraksi heksan, etil asetat, butanol, dan air, Eucheuma spinosum terdapat zat tanin. Zat tanin ini tergolong dalam senyawa fenolik. Kandungan total fenol, nilai FRAP, dan kandungan total karoten tertinggi, dihasilkan oleh fraksi etil asetat. Berdasarkan nilai ICso DPPH, nilai terbaik dihasilkan oleh fraksi butanol, diikuti oleh etil asetat Dengan demikian, fraksi etil asetat dipilih untuk digunakan pada tahapan penelitian selanjutnya, untuk diisolasi senyawanya. Bentuk senyawa aktif yang diisolasi pada fraksi etil asetat Eucheuma spinosum menggunakan teknik Kromatografi Lapis Tipis dan Kromatografi Kolom berupa padatan berwama coklat kemerahan yang larut dalam metanol. Penampakan noda pada KL T ODS setelah disemprot dengan reagen H2S04 10% dalam metanol, menghasilkan noda berwama biru. Perendaman pelat KL T yang berisi totolan sampel Eucheuma spinosum ke dalam DPPH menampakkan noda berwama kuning tua, yang menunjukkan sampel tersebut mengandung senyawa aktif antioksidan. Hal ini dibuktikan dengan hasil uji DPPH IC50 sebesar 87,97 ppm. Berdasarkan interpretasi spektrum NMR diperoleh dugaan struktur senyawa 3-(3- methoxyphenyl) propanal, dengan rumus molekul : C10H1202 yang memiliki nilai DBE 5 yang berasal dari 1 buah benzena dan 1 buah karbonil. Senyawa ini diindikasikan sebagai antioksidan, karena memiliki ikatan rangkap pada gugus aromatik dalam rumus struktumya. Adanya ikatan rangkap menyebabkan mudah bereaksi dengan senyawa lain. Berdasarkan hasil isolasi dan karakterisasi menggunakan NMR terhadap Fraksi Etil asetat Eucheuma spinosum, struktur senyawa dengan rumus molekul C10H1202 3-(3-methoxyphenyl)propanal adalah senyawa yang baru pertama diisolasi dari bahan alam alga Eucheuma spinosum. ix Lena J. SUMMARY Eucheuma cotonii and Eucheuma spinosum are marine red algae species used as food for their carrageenan content and antioxidant characteristics. Antioxidant compounds are much needed by human body, for their role in inhibiting the formation of free radicals that are the source of various diseases. Both marine algae species are so rich in bioactive compounds that they are widely cultivated to meet high market demand. As secondary metabolites component, antioxidant compounds have different activities. Presently, the studies on marine algae are still limited on the carrageenan content and pigment, either for the industry or export. Extraction of antioxidant compounds is conducted by maceration using methanol solvents at several concentrations. Isolation of compounds is undertaken through a combination of Column Chromatography separation techniques (CC) and Thin Layer Chromatography (TLC). Characterization of compounds is done through NMR. The objectives of this study are : 1) to find out the best concentration of methanol, which are able to produce the highest antioxidant activity of extracts for both fresh and dried algae Eucheuma spinosum and Eucheuma cotonii. 2) to get the product of stratified fractionation with the best solvent, which produces compounds with the best antioxidant activity. 3) to obtain compounds with antioxidant activity from selected marine algae extract through isolation with CC, TLC, and also to elucidate the compounds structure by NMR using H-NMR, CNMR, HMQC, and HMBC techniques. This study carried out in three stages, namely : (I) Extraction of antioxidant compounds from Eucheuma cotonii and Eucheuma spinosum in fresh and dry conditions using methanol at various concentrations (60, 70, and 80% ) and evaluation the water content, Total Phenol, DPPH, FRAP, and total carotenoids, (II) Testing the Phytochemical and Antioxidant Activity of selected Eucheuma sp algae through stratified fractionation using hexane solvent, ethyl acetate, butanol, with total phenols test, DPPH, FRAP, and total carotenoids, (Ill) Isolation and characterization through structure determination of antioxidant compounds algae Eucheuma sp in vitro with CC, TLC, and NMR. x ·~ .. ~ This study showed that the highest total phenol content , DPPH , and FRAP found in extractions using methanol in marine algae E. cotonii and E. spinosum (fresh and dried form) is produced by the methanol extract of 60 % in fresh E. spinosum. The highest total carotenoid content {as much as 9.40 ± 0.35 mg I g) was found in a sample of 60 % methanol extract of fresh E. cotonii, followed by 60 % methanol extract of E. spinosum (8. 73 ± 0.23 mg/g). The analysis for the four test parameters showed that fresh E. spinosum in 60 % methanol extract had better antioxidant activity compared to the other samples. This sample was used in further study. Phytochemical test showed that each extract of the hexane fraction, ethyl acetate, butanol, and water, contained tannin substance. This tannin substance belong to phenolic compounds. The highest content of total phenol, FRAP value, and total carotenes produced by ethyl acetate. Based on DPPH IC50, the best value was produced by butanol fraction, followed by ethyl acetate. Thus, the ethyl acetate fraction selected for use in the next research stage, for the isolation of the compound. Active compounds have been isolated from the ethyl acetate fraction of E. spinosum by using thin layer chromatography and columns chromatography techniques form a reddish brown solid materials that soluble in methanol. Stains on ODS TLC, when sprayed with 10% Hi$04 reagents in methanol, became a blue color stain. Soaking TLC plate containing the samples of E spinosum in to DPPH showed dark-yellow stain, which indicates that the sample contains an active compound of antioxidants. This is proved by DPPH IC50 test results as much as 87.97 ppm. Based on the interpretation of NMR spectra, the compound structure was presumed to be 3-(3-methoxyphenyl)propanal, with molecular formula : C10H1202 and has the DBE value 5 from one benzene and one carbonyl group. This compound indicated as an antioxidant, as it has a double bond at the aromatic group in the formula structure. The presence of the double bond resulted in easy reaction with other compounds. Based on the isolation and characterization using NMR for the ethyl acetate fraction of E spinosum, the structure of the compound with the molecular formula C10H1202 3-(3-methoxyphenyl)propanal is the first compounds newly isolated from natural E. spinosum algae.
